摘要
采用'醇提凝析法'从黄毛耳草中分离提纯乌索酸,用IR、MS、NMR等技术对乌索酸结构进行了表征.建立了反相高效液相色谱法测定乌索酸含量的方法,色谱柱为SymmetryShield Rp18柱(3.9 mm×150mm,5 μm);流动相为甲醇:水(88:12,V/V);流速为1.0 mL/min;二极管阵列检测器检测,检测波长为210nm;柱温为25℃;线性范围:4.5~22.5 μg;线性回归系数大于0.999 8.HPLC测定乌索酸纯度为99.82%,得率为3.65‰.该方法具有快速简便、精密度高、重现性好、线性范围宽的特点,可作为控制乌索酸质量的方法.
This paper explores adopting the method of ethanol extracting and agglutination separating to extract ursolic acid from Herba Hedyotidis Corymbosae and using spectrum to identify its' structure by IR, MS, NMR. A determination method of ursolic acid in Herba Hedyotidis Corymbosae by reversed - phase high performance liquid chromtograph was es- tablished. The chromatographic column, SymmetryShield RP18 (3.9 mm×150 mm,5μm), methanol- water (88: 12, V/V ) mobile phase with 1.0 mL/min flow rate, the detected wavelength (210 nm), and the colunm temperature(25℃) were adopted. The calibration curve of ursolic acid was linear under the content of 4.5 - 22.5μg, the correlation coefficient was over 0. 9998. The content is 99.82 % by HPLC and the gain rate is 3.65‰. The method is simple, ac- curate and has good repeatability and wider linear range. The method can be used to evaluate the quality of ursolic acid.
