摘要
目的研究实验性大鼠肝纤维化时肝星形细胞(HSC)内皮素1(ET1)、生长抑素受体2(SSR2)、血管紧张素Ⅱ受体1(AT1)、前列腺素E2受体(ER2)的mRNA表达情况及外源性白细胞介素10(IL10)干预对表达的影响。方法60只清洁级SD大鼠随机分为对照组、肝纤维化组和IL10干预组。以CCl4腹腔内注射构建肝纤维化模型,并予IL10腹腔内注射干预造模。于第7周和第11周分别随机选取一批大鼠,分离HSC并提取总RNA,以半定量RTPCR方法检测其ET1、SSR2、AT1和ER2表达。结果对照组HSC中不表达SSR2;ET1、AT1和ER2均有表达。随肝纤维化进展,ET1、SSR2和AT1表达明显增强(P<0.05),IL10干预组则减弱(P<0.05);而ER2的表达在各组HSC中无明显差异(P>0.05)。结论肝纤维化时HSC内ET1、SSR2和AT1mRNA表达增加;外源性IL10可以抑制其表达。
Objective To examine the expression of endothelin-1 (ET-1), somatostatin receptor 2 (SSR2), angiotensin Ⅱ receptor type 1 (AT1) and prostaglandin-E2 receptor(ER2) in the hepatic stellate cells of experimental rat liver fibrosis, and to investigate the effect of interleukin-10 on them. Methods Sixty clean SD rats were divided into control group, the fibrotic group and the IL-10 treated group randomly. Rat hepatic fibrosis model was induced by CCl4, and treated with IL-10 from the 3rd week. Hepatic stellate cells were isolated from these rats at the 7th and 11th weeks, respectively. The mRNA expression of ET-1, SSR2, AT1 and ER2 were detected by semi-quantitative reverse-transcription polymerase chain reaction. Results The ET- 1, AT1 and ER2 mRNA expressed in HSC of control group but the SSR2 was not found. The expression of ET- 1, SSR2 and AT1 was increased significantly with the development of liver fibrosis (P 〈 0.05), their expressions were decreased significantly (P 〈 0.05) after IL-10 treatment (P 〈 0.05). No significantly change of ER2 expression was found. Conclusions The expression of ET-1, SSR2 and AT1 in hepatic stellate cells were increased with hepatic fibrosis progress. Ectogenic IL-10 inhibits this responses.
