摘要
目的通过检测异丙酚静脉全麻诱导大鼠中枢c-jun基因的表达,了解异丙酚在大鼠中枢的可能作用位点。方法42只Wistar大鼠随机分成6组:空白对照组(C组)、小剂量异丙酚组(50mg/kg,P1组)、中等剂量异丙酚组(100mg/kg,P3组)、大剂量异丙酚组(150mg/kg,P3组)、断尾刺激组(S1组)、异丙酚作用后断尾刺激组(S2组)。免疫组织化学法(ABC法)检测JUN核蛋白,统计各组大鼠脑片的JUN免疫反应阳性神经元的分布。结果C组:JUN免疫反应阳性神经元散在分布在视上核、外侧隔核及外侧缰核等神经核团。P1、P2、P3组:JUN免疫反应阳性神经元表达较对照组明显增多,主要分布在伏核、外侧隔核、下丘脑室旁核、膝状体腹下核、背外侧膝状核、视上核、视交叉上核、视前腹侧核、弧束核、杏仁基底外侧核、丘脑室旁核、外侧缰核、海马回嗅觉小岛及乳头状核等核团,表达的阳性神经元数量与异丙酚剂量正相关。结论异丙酚在大鼠中枢神经系统有与其静脉麻醉作用相关的神经核团。
Objective To clarify the effective location of propofol in CNS, the expression of c-jun was observed after propofol-induced anesthesia in rats. Mthods Wistar rats were randomly divided into 6 groups: normal control (C), low-dose propofol group (50mg/kg, P1 ), middle-dose propofol group(100mg/kg, P2 ), high-dose propofol group(150mg/kg, P3 ), stimulation with tail broken group (S1) and propofol + stimulation with tail broken group (S2). The expressions of nucleoprotein JUN in the CNS were detected by immunohistochemisty, Results Rather weekly stained nucleoprotein JUN positive neurons were observed in supraotic nucleus, lateral septal nucleus and lateral habenular nucleus in the control group. In group P1, group P2 and group P3, it was revealed that the expressions of nucleoprotein JUN were increased significantly compared with control group, which were mainly located in accumbens nucleus, lateral septal nucleus, periventericular hypothealamic nucleus, ventral lateral geniculalaten nucleus, dorsal endapiriform nucleus, supraotic nucleus, suprachiasmatic nucleus, anteroventral preoptic nucleus, nucleus of the solitary tract, supramammillary nucleus, basolateral amygdaloid nucleus, paraventricular thalamic nucleus, LHb and islands of Calleja, ratified as a positive dose-effect relation with propofol. Conclusion Propofol anesthesia has the determined sites of action in CNS.
