摘要
本文应用随机引物扩增多态性DNA聚合酶链反应技术(RAPD-PCR)筛选出3种引物,对淡色库蚊、致乏库蚊的细胞基因组DNA进行扩增,分别扩增出不同条带数和分子大小的多态DNA图谱。两种库蚊细胞的基因组DNA扩增产物除了有相同分子大小的DNA带外,还有一半条带为各自独特的条带;用淡色库蚊成蚊作RAPD-PCR的扩增带与相应的细胞株产生的主要条带相符。实验结果表明,此技术方法简便、快速、精确,可用于蚊虫的分子分类研究。
Three primers were screened by RAPD-PCR.Different maps were obtained through amplifing the genom DNA of the mosquito cells of Culex pipiens pallens &C.pipiens fatigans with three primers(“Pz”、“Pw”、“PL”). Half of the amplified fragments are unique to one species ,and the other half fragments present in two species of Culex with the same primer.RAPD of adult mosquito DNA of Culex pipiens pallens results in the same amplified fragments as that of mosquito cells,These results indicate that the RAPD-PCR technique will be useful in studies of mosquito molecular taxonomy for its simplification、rapidness and exactness.
