摘要
应用体细胞基因敲除技术的方法和原理,通过PCR扩增获得同源短臂和长臂,将其插入骨架载体pPL622,酶切鉴定插入方向并测序确证,成功构建了人Polι基因的击靶载体PSP-PLA,其中同源短臂长980 bp,长臂长3 217 bp。
In this study, the human Pol l targeting vector pSP-PLA for somatic targeting was constructed. This targeting vector contains a short arm of 980 bp and a long arm of 3 217 bp, which are amplified by the method of PCR. The recombinants were identified by restriction digestion and verified by sequencing.
