摘要
研究了以高效液相色谱-二极管阵列法测定豆制品中的碱性嫩黄O含量的检测方法。采用Hypersil ODS-C18色谱柱,以4:6(V/V)的甲醇-0.5%乙酸铵溶液作流动相,流速1.0mL/min,用二极管阵列检测器于436nm波长下进行检测,并以保留时间和三维光谱图相似性系数进行定性。方法线性范围为0.25-50μg/mL(r=0.9999),在5.0g 样品中加入250μg标准物质的回收率及相对标准偏差(RSD)分别为95.0%(n=6)和1.5%(n=6)。
A method for determination of auramine O in legume -food by high performance, liquid chromatography-diode array detector (DAD) was studied. In this method, a hypersil ODS- C18 colume was used with methanol and 0.5% acetic-ammonium (40:60, V/V) as mobile phase. The flow rate was 1.0mL/min. A diode array detector was used and the detection wavelength was 436 nm. The calibration curve was linear over a range from 0.25μg/mL to 50.0μg/mL with a coefficient of 0.9999. Qualitative analysis was performed with retention time and DAD spectrum similarity index value. Adding 250μg auramine O standard in 5.0g sample, the recoveries were 95.0% (n=6) and relative standard deviation was 1.5%(n=6).
