摘要
比较了牡丹皮中丹皮酚的两种不同的提取方法,即水蒸气蒸馏法和乙醇渗漉法,结果乙醇渗漉法比水蒸气蒸馏法丹皮酚的提取率高.因此,进一步考察了乙醇渗漉法的乙醇浓度(75%~95%)、浸泡时间(12~36h)、渗漉速度(2~6ml/min)、乙醇用量(6~10倍)对牡丹皮中丹皮酚提取的影响,利用正交实验设计(4因素3水平)获取9个不同实验条件,并用HLPC法检测丹皮酚,对实验结果分析并结合实际生产得出结论.乙醇渗漉法的最佳条件:乙醇浓度75%、浸泡时间12h、渗漉速度6ml/min、收集相当药材8倍量的体积,丹皮酚的提取率为最高95%.乙醇渗漉法比水蒸气蒸馏法提取丹皮酚,工艺流程简单,可操纵性强,得率高.确定的参数可为以后的生产提供参考.
The methods of water vapour distillation and percolation were compared on the extraction ratio of the paeonal in cotex moutan . As a result, the extraction ratio of percolation was better. Farther,effects of the concentration of ethanol, the time of soak, the rate of percolation and the volume of solventon on the extraction ratio of the paeonal were studied in the method of percolation . The experimental conditions were obtained by positive coross trial . The best parameter of technology showed that the concentration of ethanol 75 % , the time of soak 12h , the rate of percolation 6ml/min,and the volume of solventon 8 times, the extraction ratio of the paeonal was about 95 %.
