摘要 目的探讨丁酸钠(N aB)对人卵巢上皮癌细胞株3AO增殖和凋亡的影响及其作用机制。方法用不同浓度N aB对3AO细胞作用不同时间,观察其对3AO细胞增殖活性的影响,分析细胞周期、DNA含量和细胞凋亡率的变化。结果随培养时间延长,N aB浓度≥4mm o l/L时,3AO细胞皱缩,数量渐减;同一N aB浓度下,3AO细胞增殖抑制率随作用时间的延长有增加趋势;N aB浓度为4mm o l/L、8mm o l/L、12mm o l/L时,对3AO细胞增殖有明显抑制作用;N aB作用72h,随浓度升高,增殖细胞核抗原(PCNA)阳性细胞百分率逐渐下降,与对照组相比差异有显著性,P<0.05。N aB作用24h开始出现细胞周期阻滞,随浓度增高,G1期细胞增多,S期细胞减少,作用48h,出现细胞凋亡峰;增加N aB浓度或相同浓度增加作用时间,凋亡峰增高。结论N aB呈剂量、时间依赖模式影响卵巢上皮癌细胞株3AO的体外增殖,可能通过G1期阻滞诱导卵巢上皮癌细胞凋亡。 Objective :To investigate the influence of sodium butyrate(NaB)to ovarian epithelial cancer cell line 3AO and detect the probable mechanism. Methods :NaB of different concentration was added to cultured 3AO cell in vitro. The proliferation activities of 3AO ceils were measured. Cell apoptosis were studied. Results :As cultured time goes by, 3AO ceils cultured with NaB decreased both in quality and quantity according to the concentration of NaB. The proliferation of 3AO cells was inhibited by NaB in a does-time-dependent manner. Being cultured with NaB for 72h,with the rose of drug concentration,the PCNA positive cell reduced. G1 cells were arrested by NaB in 24h,apoptotic peak appeared at 48h,and the peak went higher as drug concentration rose. While at the same concentration,the peak rose as the treating time prolonged. Conclusions:NaB at certain concentration can inhibit proliferation and induce apoptosis in 3AO cells. G1 cells arrest may be involved in the mechanism of apoptosis.
