南虎凝胶剂抗炎镇痛作用及其作用机制

摘要 目的:评价南虎凝胶剂的抗炎镇痛作用,揭示其抗炎镇痛作用机制.方法:用二甲苯致小鼠耳肿胀、角叉菜胶致大鼠足爪肿胀模型,考察南虎凝胶剂的抗炎作用;热板法观察南虎凝胶剂的镇痛作用.用大鼠IL-1βELISA试剂盒和大鼠TNF-αELISA试剂盒检测角叉菜胶致炎大鼠足跖肿胀组织中IL-1与TNF-α含量,了解南虎凝胶剂对炎性组织中IL-1及TNF-α的影响,揭示其抗炎镇痛作用机制.结果:南虎凝胶剂外用给药,可明显减轻小鼠耳肿胀度,降低角叉菜胶所致大鼠足爪肿胀,明显延长热刺激小鼠痛阈值,降低炎性组织提取液中IL-1及TNF-α含量.结论:南虎凝胶剂对炎症反应具有明显的抑制作用和中枢镇痛作用,其抗炎镇痛作用机制可能与其抑制IL-1与TNF-α产生有关. AIM: To estimate the anti-inflammatory and analgesic effects of Nanhu Gelata and to study its mechanism. METHODS: Xylene-induced mouse ear edema models and carrageenan-induced rat paw edema models were used to study the anti-inflammatory effects, Hot-plate test in mice was used to study the analgesic effects, The contents of IL-1 and TNF-α in the carrageenan-induced rat paw edema tissues were measured using rat IL-1β ELISA and TNF-α ELISA kits to explore the mechanism of its anti-inflammatory and analgesic effects, RESULTS: Nanhu Gelata significantly suppressed xylene-induced mouse ear edema and carrageenan-induced rat paw edema, The pain threshold of hot-plate test mice was remarkably improved by Nanhu Gelata. The content of IL-1 and TNF-α in the inflammatory tissue extracts significantly decreased. CONCLUSION: Nanhu Gelata has some significant anti-inflammatory effects on acute inflammatory reactions, Nanhu Gelata significantly re- lieves pain reactions induced by heat, The mechanism of anti-inflammatory and analgesic effects of Nanhu Gelata may be related to its effect of decreasing the IL-1 and TNF-α content in inflammatory tissues.
