摘要 目的研究抵抗素基因外显子4第1308的位点核苷酸G/A变异与武汉地区2型糖尿病和肥胖者发病风险的关系。方法应用聚合酶链反应-限制性片段长度多态性方法检测武汉地区汉族160例抵抗素基因外显子4第1308位点的G/A变异。结果2型糖尿病组和肥胖组的GG、AA基因型及G/A等位基因频率与正常组比较,均有显著性差异(P<0.05)。GG基因型携带者使个体患肥胖和糖尿病的危险性分别增加1.33倍和2.22倍,将G等位基因视为暴露因素,其使个体患肥胖以及糖尿病的危险性分别增加1.12倍和1.80倍。结论抵抗素基因外显子4第1308位点核苷酸多态性与2型糖尿病和肥胖的易感性相关,等位基因G可能是由肥胖致2型糖尿病的一个重要的相关因素。 Objective To investigate the relationship between 1308G/A variant of the fourth exon of resistin gene and the risk of type 2 diabetes mellitus and obesity in Wuhan district. Method The study included 160 subjects in Wuhan Hans. The genotypes of 1308G/A variant of the resistin gene were determined with polymerase chain reaction-restriction fragment length polymorphism assay. Results There was significant difference of GG ,AA genotypes and G/A alleles frequencies between patients with diabetes mellitus or obesity and health subjects ( P 〈 0.05 ). The risk of GG genotype for diabetes mellitus and obesity was 1.33 times and 2.22 times higher than that in A/G or AA genotype, respectively. The individuals with allele G had 1.12-or 1.18-fold increases of the risk for diabetes mellitus and obesity. Conclusion The resistin gene (G1308A) single nucleotide polymorphism was associated with susceptibility to diabetes mellitus and obesity in Wuhan Hans. The allele G has an increased risk of diabetes mellitus and obesity.
