非小细胞肺癌中p14^(ARF)基因的表达及意义

摘要 ①目的检测p14ARF基因在非小细胞肺癌(NSCLC)中表达情况,并探讨其在NSCLC发生、发展过程中的作用。②方法应用蛋白印迹方法检测了28例NSCLC组织标本p14ARF蛋白的表达水平。③结果肺鳞癌和肺腺癌中p14ARF蛋白阳性率分别为61.5%和80.0%,与癌旁正常组织(100%)比较差异有显著性(χ2=12.27、6.02,P<0.05);p14ARF表达异常与肿瘤分期、组织类型、分化程度、淋巴结转移情况无显著相关性;p14ARF蛋白在Ⅰ期病例中强阳性占阳性比例为62.5%,在Ⅲ期病例中强阳性占阳性比例为16.0%。④结论p14ARF基因在NSCLC发生、发展过程中起重要作用,特别在肿瘤早期可能起更为重要的作用。 Objective To study the role of p14^ARE gene in oncogenesis of non-small cell lung cancer (NSCLC) and its expression in the disease. Methods The expression of p14^ARF gene was detected in 28 cases of NSCLC by western blot. Results The positive rate of p14^ARF gene expression of squamous carcinoma and adenocarcinoma in NSCLC was 61.5 %and 80.0% respectively, which was significantly different from that in normal tissues adjacent to cancer (100% )(x^2=12.27,6.02;P〈0.05). The expression level of p14^ARF gene was not significantly related to histological classification, TNM stage, cancer cell differentiation and lymph node involvement in NSCLC. The rate of strong staining (+++) cases to all the positive cases was 62. 5% in stage Ⅰ and 16.0% in stage Ⅲ. Conclusion The p14^ARF gene plays an important role in the occurrence and development of NSCLC, especially in the early stage of the disease.
