青紫薯色素对自然衰老大鼠体内抗氧化能力的影响 被引量:8

摘要 ①目的探讨青紫薯色素对自然衰老大鼠体内抗氧化能力的影响.②方法以健康青年Wistar大鼠为正常对照组,50只自然衰老Wistar大鼠分为模型组,青紫薯色素高、中、低剂量(1 500、150、15 mg/kg)组和维生素E(2 mg/kg)组.正常对照组和模型组给予生理盐水20 mL/kg,其他各组每日灌胃1次,每次3 mL,连续给药30 d.应用生化方法检测青紫薯色素对自然衰老Wistar大鼠体内两种脏器(脑、心脏)组织匀浆的超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-px)和总抗氧化能力(T-AOC)的影响,并探讨青紫薯色素剂量与三种抗氧化指标之间的相关关系.③结果自然衰老大鼠脑、心脏组织匀浆中SOD、GSH-px和T-AOC活性均显著降低,青紫薯色素可以提高衰老大鼠组织匀浆SOD、GSH-px的活性,增强衰老大鼠机体的T-AOC,与模型组比较,差异具有显著统计学意义(F=9.05~44.09,q=2.86~12.10,P<0.05).3种抗氧化指标中,脑组织中SOD、GSH-px和T-AOC水平随青紫薯色素剂量的增加而增高(r=0.87~0.93,P<0.05);而心脏组织中SOD、GSH-px和T-AOC水平与青紫薯色素的剂量不呈正相关关系(r=0.43~0.51,P>0.05).④结论青紫薯色素可以通过有效调节自然衰老大鼠体内的氧化水平,延缓衰老进程,剂量依赖性地提高脑组织的抗氧化能力. Objective To investigate the effect of sweet-potato anthocyanins on oxidant level in aged rats. Methods Fifty natural aging Wistar rats were randomly divided into 5 groups: model group, low dose, moderate-dose, high dose sweet-potato anthocyanins groups (1 500, 150, 15 mg/kg, respectively) and vitamin E (2 mg/kg) group. Young healthy Wistar rats were served as normal control group (20 mL/kg sodium chloride) with intragastric administration 3 mL/d for 30 d. The activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-px) and total antioxidant capacity (T-AOC) in brain and heart in aged rats were detected using biochemical method. The correlation between the organs and antioxidant species were investigated. Resuits Decreased SOD, GSH-px and T-AOC were observed in aged rats. Administration of sweet-potato anthocyanins significantly increased the activity of SOD, GSH-px and T-AOC in the brain tested in aged rats (F=9.05-44.09, q=2. 86-12.10, P〈0.05) in a dose-dependent manner. Conclusion Correlated antioxidant activity of the organs in aged rats is in accordance with the normal aging process. The sweet-potato anthocyanins can be used to compensate the decline in T-AOC and the activities of antioxidant enzymes and thereby to inhibit the aging process.
