摘要
目的:观察淫羊藿总黄酮(epimediumpubescensflavonoids,EF)能否抑制己烯雌酚(diethyl-stibestrol,DES)刺激子宫不良反应,及对DES抗骨质疏松作用的影响。方法:50只4个月龄SD雌性大鼠随机分为5组:假手术组(Sham组);去卵巢组(OVX组);己烯雌酚组(DES组);淫羊藿总黄酮组(EF组);联合用药组(EF+DES组)。实验12周,通过测量子宫内膜厚度及子宫湿重分析用药对子宫的刺激作用,测量骨小梁面积百分率(%Tb.Ar)、荧光周长百分数(%L.Pm)及破骨细胞数量(Oc.N)反映骨量、骨形成及骨吸收的变化。结果:DES组与OVX组比,子宫湿重及内膜厚度都显著增加,%Tb.Ar增加,%L.Pm、Oc.N减少;EF组与OVX组比,子宫湿重及内膜厚度、%Tb.Ar、Oc.N均无显著性差异;EF+DES与DES组比,内膜厚度显著降低,%Tb.Ar增加;与假手术组比各参数无显著性差异。结论:淫羊藿总黄酮可有效抑制己烯雌酚对子宫内膜刺激作用并增强其防治骨质疏松作用。
Objective: To observe the effects of combined medication of epimedium pubescens flavonoids (EF) and diethylstilbestrol (DES) on uterus and bones in ovariectomized (OVX) rats. Methods:Fifty female SD rats were divided into 5 groups:Sham-OVX group,OVX group,DES group,EF group and combined medication of EF and DES group (EF+ DES group). After an experiment period of 12 weeks, the side-effects on uterus was analyzed by measuring the wet weight of uterus and the endometrium epithelial thickness(EET) and the changes of bone mass with quantitative bone histomorphometry were observed. Results: The percent of bone trabecala area ( % Tb. Ar), uterine wet weight and EET in DES group were significantly increased as compared with OVX group. EF+ DES group showed increase in% Tb. Ar and decrease in uterine weight and EET,as compared with DES group. There was no significant difference between EF+ DES group and shamOVX group. Conclusion:EF could inhibit the DES's stimulative side effect on uterus and increase its anti-osteoporosis effects.
