内源性成骨因子在兔硬脑膜再生骨中的表达

The Expression of Endogenous Osteoinductive Factors in Dura Mater Osteogenesis of Rabbits

摘要 目的:用外源性重组人骨形态发生蛋白-2(rhBMP-2)诱导兔结论:硬脑膜再生骨,研究内源性成骨因子骨形态发生蛋白-2(BMP-2)和转化生长因子-β1(TGF-β1)的表达。方法:在诱导区加入rhBMP-2,应用免疫组化方法检测硬脑膜内源性BMP-2和TGF-β1不同天数的表达。结果:术后早期聚集的大量间充质细胞即出现表达BMP-2和TGF-β1,且随着间充质细胞分化为前软骨细胞并不断成熟,两因子的表达水平迅速提高。BMP-2在术后11 d表达最活跃,之后开始下降,而TGF-β1表达高峰出现在术后17 d。结论:外源性rhBMP-2在诱导颅骨再生过程中,能增强内源性BMP-2及TGF-β1在硬脑膜的表达,且BMP-2表达高峰早于TGF-β1。 Objective: To study the expression of endogenous bone morphogenetie protein-2(BMP-2) and transforming growth factor-β1 (TGF-β1)during rabbits' dura mater osteogenesis induced by recombinant human bone morphogenetie protein-2(rhBMP-2). Method: The immunohistochemical method was used to detect the expression of endogenous BMP-2 and TGF-β1 in different days after combined with rhBMP-2 in inducement area. Results: With positive expression of endogenous BMP-2 and TGF-β1, a lot of mesenehymal cells aggregated in dura mater in the early stage after operation, and the expression level of BMP-2 and TGF-β1 increased quickly with the differentiation of mesenehymal cells. BMP-2 expression was in the highest on day 11, while TGF-β1 on day 17. Conclusion: During osteogenesis induced by rhBMP-2, exogenous rhBMP-2 can reinforce the expression of endogenous BMP-2 and TGF-β1, and the expression peak of BMP-2 is earher.
