ANALYSIS OF CELLUAR IMMUNE FUNCTIONS IN THE PROCESS OF RENAL FIBROSIS IN A RAT ADR MODEL

摘要 目的探讨肾纤维化大鼠细胞免疫功能状态。方法建立阿霉素肾病大鼠模型,3H-TdR掺入法检测脾细胞增殖能力,流式细胞术检测外周血T细胞亚群、NKT细胞及Th1/Th2细胞。结果肾纤维化大鼠脾细胞对Con A诱导的增殖能力明显下降(P<0.05),T亚群细胞的比例发生改变,随着肾纤维化的进程,CD3+、CD4+所占比例降低,而CD3+、CD8+细胞增多,CD4+/CD8+逐步下降(P<0.05)。外周血CD3+、CD56+细胞却呈上升趋势,机体处于明显的Th2偏移状态。结论肾纤维化大鼠出现明显的细胞免疫功能紊乱。 Objective To investigate the cellUar immune functions in the process of renal fibrosis. Methods The rat model of ADR inducing renal fibrosis was established; the proliferation of spleen lymphocyte was examined by 3H-TdR incorporation; T subset, NKT cell and Th1/Th2 in PBMC were detected by flow cytometry. Results Compared with control group, the proliferation of the Con A * . inducing spleen lymphocyte descend significantly along with the process of renal fibrosis. Unlike the declined CD3^+, CD4^+ cells, the proportion of CD3^+, CD8^+ cells increased. There was a sharp increase of CD3^+ , CD56^+ ; the rate of CD^4 + IFN - γ +/CD4 + IL-4^+ significantly decreased. Conclusion The celluar immune function was disturbed in the rat ADR model which may contribute to speed the outcome of the renal fibrosis.
