成人急性白血病淋巴细胞HSP70蛋白及mRNA的表达研究

Research of HSP70 and mRNA Expression in Lymphocyte of Adult Acute Leukemia

研究热休克蛋白70(HSP70)在成人急性白血病淋巴细胞中的表达。用W estern blot法检测化疗前、后及正常人HSP70的蛋白表达,RT-PCR法检测其HSP70mRNA的表达。化疗前成人急性白血病淋巴细胞HSP70蛋白表达明显低于正常人(P<0.05);其HSP70mRNA表达的灰度值为(0.112±0.005),明显低于正常(人0.198±0.002)(P<0.05);而化疗后,成人急性白血病淋巴细胞HSP70及其mRNA(0.287±0.001)明显高于正常人(0.198±0.002)(P<0.05)。HSP70在成人急性淋巴细胞白血病淋巴细胞中高表达,对癌细胞起保护作用。

To detect the expression of heat shock protein 70 （HSPT0） and its mRNA in lymphocyte of adult acute leukemia, HSP70 was determined before chemotherapy, after themotherapy and in normal by Western blot. HSP70 mRNA was determined by RT-PCR. The espression of HSP70 in adult acute leukemia lymphocyte was found to be much lower than normal before chemotherapy （P 〈 0.05） ;the gray scale value（GS） （0. 112 ± 0. 005） was much lower than normal[ 0. 198 ± 0. 002 （ P 〈 0.05 ） ], too. Yet after themotherapy, the expression of HSP70 and mRNA was much higher in adult acute leukemia lymphocyte than normal [0. 198 ±0. 002 （P 〈 0.05） ]. There is close relationship between adult acute leukemia cells and HSP70, which protects cancer cells.
