摘要 目的:建立用高效液相法(HPLC)测定血清中文拉法辛浓度的方法.方法:HPLC测定,流动相为甲醇:乙腈:水:1 mol/L醋酸铵:5 mol/L氨水(350:50:100:5:1)的混合液体,pH 8.7,流速1 ml/min,ODScol柱,紫外检测波长229 nm,以外标法定量.结果:线性范围25~700ng·ml-1,最低检测限2 ng·ml-1,平均提取回收率96.59%,日内和日间精密度平均分别为3.44%和3.77%.结论:用HPLC可以较好的对血清中文拉法辛进行定性和定量. Objective:To establish a HPLC assay method for the determination of Venlafaxine level in human serum. Methods: Using a mixture liquid of Acetonitrile : Methanol : Water : 1 mol/L - AmmoniumAcetate : 5mol/L - NH4OH (50 : 350 : 100 : 5 : 1 ) as mobile phase,pH8.7, flow speed was 1ml/min, ODScolumn, the wave length of detection was229nm, Venlafaxine level was determined by means of external standard method. Results : A linearity was obtained from 25 - 700ngoml - 1 of Venlafaxine level in serum with a good correlation ( r = :0. 998 ). Detection limit was 2ngoml - 1 . The average extraction recovery was 96.59 % . The coefficients Of inter - day and intra - day variations were 3.44 % and3.77 % respectively. Conclusions : Using HPLC can detect Venlafaxine in serum well.
