摘要
目的探讨心气虚证心肌细胞模型β受体、ET和NOS基因表达的改变。方法采取“培养心肌细胞缺氧再给氧损伤方法”建立心气虚细胞模型,设对照组和模型组,采用逆转录聚合酶链反应(RT-PCR)基因表达检测心气虚细胞模型β受体、ET和NOS基因表达情况。结果心气虚细胞模型与对照组的β1受体mRNA、ET mRNA和NOS mRNA改变情况相比较,差异非常显著(P<0.01)。结论心气虚证的分子基因水平的病理生理学基础与β1受体、ET和NOS基因表达有关。
Objective:To study the changes of β-receptor,ET and NOS genes expression in deficiency of heart qi syndrome (DHQS) cell model.Methods:The DHQS cell model was set up by using cultured cardiac muscle cells which were damaged by hypoxiatingthen replenishing its oxygen supply.The RT-PCR gene expression technique was both performed on the experimental cell model and the control group.The result of the gene expression on β-receptor,ET and NOS were measured.Results:Comparing the gene expression of β-receptor mRNA,ET mRNA and NOS mRNA between DHQS cell model group and the control group,the difference was very significant(P<0.01).Conclusion:The pathophysiological base of DHQS molecular genetics was related to β-receptor,ET and NOS genes expression.
