液相色谱法测定大白鼠中枢神经系统受损时释放的氨基酸量

摘要 提出以NDA为衍生化试剂,利用柱前衍生化技术,简便、准确和灵敏地测定氨基酸的方法.色谱柱为ODS柱,萤光检测器,梯度洗脱,洗脱液A为10%THF和90%磷酸盐缓冲液,洗脱液B为55%乙腈、10%甲醇和35%磷酸盐缓冲液。该方法可用来测定活体老鼠脊髓遭受超氧游离基损伤时释放的氨基酸量. A simple,accurate and sensitive method for determination of amine acids released from injured spinal cord of rats by HPLC is described.The pre-column derivatizator is NDA,C18 column(3μm)of 150 min×4. 6 mm is used.Mobile phase A was 10% THF and 90% 0.05 mol/L potassium phosphate buffer adjusted to pH 5.82,mobile phase B was 55% acetonitrile, 10% methanol,and 35% potassium phosphate buffer.Microdialyser was used to administer paraquat into the spinal cord of rats to determine the released quantity of AA in injured CNS caused by superoxide anino.
