17-β雌二醇对卵巢去势大鼠延髓神经元超微结构的影响

The Effect of 17-β Estradiol on Neuron Ultrastructure of the Medulla Oblongata in Ovariectomized Rats

摘要 目的观察雌激素对卵巢去势大鼠延髓神经元超微结构的影响。方法30只雌性成年SD大鼠随机分为去卵巢组(A组)、雌二醇组(B组)和假手术对照组(C组),A组和B组动物卵巢切除后10天超分别皮下注射生理盐水0.1 m l/d、17-β雌二醇20μg/kg/d。C组动物,只切开腹膜,不切除卵巢,手术后10天起分别皮下注射生理盐水0.1 m l/d。各组动物连续用药6周后,观察结果。结果血清雌二醇的浓度A组明显低于C组(P<0.01),B组明显高于A组(P<0.01),而B组和C组之间比较无差异。A组动物延髓孤束核神经元和神经胶质细胞超微结构有明显的损伤表现,细胞核形状不规则,核周间隙局部明显增宽,胞浆内有空泡,线粒体肿胀,嵴断裂;神经纤维有脱髓鞘,髓鞘板层分离,有空泡,线粒体肿胀。B组动物,在应用雌激素替代治疗后,延髓神经元的损伤表现明显减轻。C组神经元形态结构正常。结论雌激素对去势大鼠延髓神经元有明显地保护作用。 Objective To study the effect of 17-β estradiol on neuron uhrastructure of solitary nucleus in ovariectomized rats. Methods 30 adult female rats were randomly divided into ovariectomy group (group A), estradiol group (group B ) and sham-ovariectomy group( group C). The rats of group A and group B were bilaterally ovariectomized and treated with normal saline(0. 1 ml/d) and 17-β estradiol(20 μg/kg/d) resptecively. The rats of group C were just injected with normal saline(0.1 ml/d) for 6 weeks without orariectomy. Results The level of serum estradiol in group A was significantly decreased compared with group C( P 〈0.01 ) ,and also lower than that in group B( P 〈0.01 ) without obvious difference between B and C groups. The uhrastructures of neurons and neurogliocytes in solitary nucleus were abnormal in group A with atypical nucleus shape, enlarged perinuclear cisternae, vacuolated cytoplasm, dilatated mitochondria and breaked ridge and myelinoclasis of nerve fibers. These changes were much milder in group B than in group A,with normal uhrastructure in group C. Conclusion 17-β estradiol has effect on protection of neurons of medulla oblongata in ovariectomized rats.
