新型胆碱酯酶抑制剂(NAI)对AD大鼠海马结构和红细胞膜AChE的抑制作用 被引量:1

摘要 目的探讨新型胆碱酯酶抑制剂(NAI)对阿尔茨海默病(AD)大鼠海马结构神经元胞浆和血液红细胞膜的抑制作用,为AD病的临床治疗提供新的基础研究资料。方法选用48只W istar雄性大鼠,随机分为4组(n=12):NAI组、石衫碱甲(Hup)组、AD组、空白(B lank)组。将前3组大鼠切断穹窿-海马伞制作AD动物模型。2周后静脉给予NAI和Hup,采用分光光度计对各组大鼠海马结构神经元胞浆和红细胞膜分别进行AChE酶活性测定。结果AD海马结构神经元胞浆和红细胞膜的AChE活性均较B lank组明显增高(P<0.01);NAI组上述两个部位的AChE活性较AD组显著降低(P<0.01),Hup组与AD组比较其活性也降低(P<0.05)。并且NAI组上述两个部位的AChE活性均较Hup组降低(P<0.05)。结论NAI和Hup对AD大鼠海马结构神经元胞浆和血液红细胞膜AChE均具有较强的抑制作用,NAI抑制作用强于Hup。 Objective To investigate the biochemical effects of a novel AChE inhibitor(NAI) on the neuron plasma of the hippocampal formation and the erythrocyte membrane of Alzheimer' s Disease (AD) rats and provide basic reliable scientific data for AD clinical treatment. Methods 48 male Wistar rats of 3 - 5 months were randomly divided into 4 groups : NAI group, Hup group, AD group and Blank group. Fimbria-fomix (FF) of the first three groups was transected to make AD models. 2 weeks after operation, two groups received intravenous injection of NAI and huptrzin A(Hup) respectively,adapting the spectrophometry to test the AChE activity in hippocampal formation and erythrocyte membrane of each group. Results The AChE activities of the neuron plasma in hippocampal formation and the erythrocyte membrane of the AD group were increased dramatically comparing to the blank group ( P 〈 0. 01 ). However,the NAI and Hup groups performed the contrary factor compared with the AD group,while the effect of NAI( P 〈 0. 01 ) is more obvious than that of Hup( P 〈 0.05 ). Within these two groups ,the characteristic of NAI group was reduced when taking the Hup group into consideration ( P 〈 0.05 ). Condusion Both of NAI and Hup play an important role in reducing the AChE level ,the inhibitive function of NAI is much stronger than that of Hup.
