摘要
目的研究转化生长因子-β1(TGF-β1)抗体和血小板生长因子(PDGF)抗体防治肝纤维化和门脉高压的分子机制,并探讨二者的关系。方法分别用于TGF-β1抗体和PDGF抗体处理大鼠肝星状细胞24h后,应用膜片钳全细胞技术观察细胞L型钙通道。利用正交设计方法研究不同浓度TGF-β1抗体、PDGF抗体及其作用的先后顺序和二者作用的时间间隔对肝星状细胞L型钙通道的影响。结果不同浓度TGF-β1抗体、PDGF抗体及其作用的前后顺序对细胞L型钙电流的影响有显著性(P<0.05),两因素作用的时间间隔对结果影响无显著性(P>0.05)。结论TGF-β1抗体和PDGF抗体均具有防治肝纤维化和门脉高压作用,其机理可能与其调节肝星状细胞L型钙通道有关。
Objective To study the molecular mechanism of transforming growth factor-β1 (TGF-β1 ) antibody and platelet derived growth factor(PDGF) antibody against liver fibrosis and portal hypertension, moreover explore their relationship. Methods Hepatic stellate cells(HSCs) from rats were treated with TGF-β1 antibody and PDGF antibody for 24 hours respectively and L-type calcium channels in cultured hepatic stellate cells were observed by conventional whole-cell mode of patch clamp technique. Orthogonal design was used to study the change of L-type calcium channels in hepatic stellate cells treated with TGF-β1 antibody and PDGF antibody, two antibodies action order and time span. Results There were significantly differences in effect on L-type calcium channels current from different concentrations of TGF-β1 antibody, PDGF antibody and their order( P 〈 0.05), but no significance in their time span( P 〉 0.05 ). TGF-β1 antibody and PDGF antibody within 5 - 10 μg/ml sharply increased the L-type calcium channels current( P 〈 0.05). Conclusions The mechanism of TGF-β1 antibody and PDGF antibody against liver fibrosis and portal hypertension might be related to the inhibition of L-type calcium channels in hepatic stellate cells.
