大鼠骨髓基质细胞体外诱导分化为神经细胞的研究

摘要 ①目的探讨大鼠骨髓基质细胞体外向神经细胞分化的特性。②方法大鼠的骨髓基质细胞传代培养4代后,以成纤维细胞生长因子(bFGF)及β-巯基乙醇预诱导24小时,再以β-巯基乙醇、二甲基亚砜和3-叔丁基4-羟基茴香醚联合进行正式诱导6小时。诱导后的细胞采用免疫组化法进行鉴定。③结果诱导后大鼠骨髓基质细胞神经元特异性烯醇化酶(NSE)和胶质纤维酸性蛋白(GFAP)的表达率分别为84.13%和4.27%。④结论β-巯基乙醇、二甲基亚砜和3-叔丁基4-羟基茴香醚可以促进大鼠骨髓基质细胞向神经元和神经胶质细胞分化。 Objective To investigate the feasibility in vitro inducing differentiation of bone marrow stromal cells(BMSCs) into neural ceils,and to offer reference for tile application of BMSCs in nem^science. Method The bone marrow stromal cells from Wistar rats were cultured for 4 generation. The BMSCs were incubated with L - DMEM containing 20% FBS , 10ng/ml bFGF aim 1 mM BME for 24 hours. Then the media were replaced by L - DMEM with 20% FBS, 10mM BME, 200μM BHA and 2% DMSO for 5 hours. The morphological changes of the ceils were observed under phase contrast microscope. The ceils were stained immunocytochemically with antiNSE and GFAP antibodies respectively. Results BMSCS could be induced to neural cells expressing neural ceils specific markers NSE and GFAP,the rate is 84.13% and 4.27%. Conclusions BMSCS could be induced to neural ceils mostly consisted of neuron like cells by BME, BHA and DMSO.
