摘要
采用PCR-RFLP方法对金水乌鸡、武定鸡、湖北红鸡、青脚麻鸡、矮黄鸡和隐性白羽肉鸡6个品种共计360只鸡的FABP基因进行多态性分析,选用HhaⅠ、HaeⅢ、HinfⅠ、BamHⅠ、EcoRⅠ和PstⅠ6种限制性内切酶对PCR产物进行单酶切,结果发现HhaⅠ-RFLP具有多态性,有AA和AB两种基因型,该基因座频率分布经独立性χ2检验,结果表明,蛋用品种湖北红鸡与其他品种间存在显著或极显著差异,群体中B基因频率占明显优势;矮黄鸡与隐性白羽肉鸡2个肉用品种鸡与其他品种间也存在显著或极显著差异,这2个品种的优势基因为A基因。
In this article,the polymerase chain reaction-restietion fragment length polymorphisms (PCR-RFLPs)of FABP gene in 360 chickens from 6 chickens breeds including Jinshui silk ehieken,Wuding chieken,Hubei red egg chieken,Qinjiaoma chicken,orange dwarf chicken and white recessive chicken were investigated. Six restrictions of Hha Ⅰ ,Hoe Ⅲ ,Hinf Ⅰ ,Barn H Ⅰ ,Eco R Ⅰ and Pst Ⅰ were used to digeste the production of PCR respectively.It was demonstrated that difterent genotypes (AA and AB) of FABP gene had been detected for Hha Ⅰ site.The result of Chi Square(x^2) test showed that the difference of genetype frequency between Hubei red egg chicken,orange dwarf chicken or white reeessive chicken and the other 5 chicken breeds was significant (P〈0.05) or greatly significant (P〈0.01).The B gene was the superior gene in Hubei red egg chicken and in the two broiler chicken breeds. The frequency of A gene was greatly higher than B gene.
