摘要
目的:研究地心芩总黄酮(Melastomadodecandrumflavonoids,MDF)清除氧自由基与抗脂质过氧化作用。方法:以黄嘌吟-黄嘌呤氧化酶系统产生的超氧阴离子自由基,研究MDF对氧自由基的清除作用;分别以NAPDH-维生素C和Fe2+-半胱氨酸系统诱发的肝线粒体脂质过氧化的产物,研究MDF抗脂质过氧化的抑制作用。结果:MDF能有效地清除氧自由基,其IC50为46·4mg/L;能抑制小鼠肝匀浆自氧化,能有效地抑制NAPDH-维生素C和Fe2+-半胱氨酸系统诱发的肝线粒体脂质过氧化作用,对肝线粒体有保护作用,并呈浓度依赖作用。结论:MDF对氧自由基有清除作用,并能预防性对抗超氧阴离子自由基引起的氧化性损伤,对肝线粒体的氧化性损伤有保护作用。
Objective:To study the effects of Melastoma dodecandrum flavonoids(MDF) on scavenging oxygen free radicals and inhibiting lipid-peroxidation of mice liver mitochondria in vitro. Methods. The lipid-peroxidation (malondialdehyde fromation)in mice liver mitochondria induced by NAPDH - Vit C 和 Fe^2+ - Cys, and the superoxie anion radical production in xanthine-xanthine oxidase system in vitro were evaluated by TBA spectrometric methods. Results : MDF could significantly inhibit the superoxide anion radical, IC50 is 46.4mg/L, and the lipid peroxidation of mice liver homogenate and mitochondria induced by auto-oxidation and induced-oxidation. The hibition exhibited concentration-dependent manner. Conclusion : It indicated that MDF could scavenge oxygen free radicals and was capable of prophylactically antagonizing against the oxidative injury by superoxide anion.
