摘要 目的研究藏药旺拉有效活性成分CE对D-半乳糖(D-Gal)合并亚硝酸钠(NaNO2)建造的衰老小鼠免疫器官指数以及神经系统病理改变的影响。方法采用腹腔注射D-Gal(按体重120 mg/kg)和NaNO2(按体重90mg/kg)的方法建立衰老模型,以称重法测定胸腺指数和脾脏指数,经HE染色后观察小鼠海马部位的病理改变,用免疫组化染色方法测定海马内APP、Tau和NT-3的阳性细胞数,并进行灰度扫描以反映染色的深浅。结果D-Gal合并NaNO2建造的衰老小鼠免疫器官指数下降,而CE(按体重2.5、5、10 mg/kg,口服)可减轻免疫器官指数的下降程度。衰老小鼠脑中APP、Tau阳性神经元数量明显增加,且NT-3阳性神经元数量也减少,而CE(按体重2.5、5、10 mg/kg,口服)可抑制APP、Tau阳性神经元增多且可使NT-3的阳性细胞数增加。结论藏药旺拉有效成分CE可明显改善D-Gal合并NaNO2建造的衰老小鼠免疫器官指数和神经系统病理改变。 Objective To study the anti-aging effects of CE, an effective extract isolated from a Tibetan traditional herbal medicine, on amnestic mice induced by D-galactose and NaNO2. Methods The aging mice were induced by consecutive ip injection of D-galactose (120 mg/kg body weight) and NaNO2 (90 mg/kg body weight). The thymus index and spleen index were calculated to evaluate the function of immunosystem. The HE staining of hippocampus sections was performed to reveal neuron morphological changes. The expressions of APP,Tau and NT-3 protein in brain tissue were studied by immunohistochemical staining. Results Compared with the D-galactose and NaNO2 group, CE (2.5,5,10 mg/kg body weight, po) increased the index of thymus and spleen remarkably. Combined injection of D-galactose and NaNO2 resulted in the number of APP, Tau positive cells increased remarkably and those of NT-3 decreased. CE (2.5,5,10 mg/kg body weight, po) could rectify these changes. Conclusions These results indicate that CE could exert protect effects on the aging mice of consecutive injection of D-galactose and NaNO2.
