糖尿病大鼠下颌下腺内瘦素和瘦素受体表达变化 被引量:2

Leptin and its receptor expression in the submandibular glands of streptozotocin-induced diabetic rats

摘要 目的观察糖尿病大鼠下颌下腺内瘦素和瘦素受体表达的变化。方法SD雄性大鼠24只,随机分为实验组12只(4、12周各6只)及对照组12只。用链脲佐菌素复制糖尿病动物模型,分别于4、12周后测体重和血糖,应用HE和免疫组织化学SABC法,观察组织学变化并检测瘦素及其受体表达。结果与对照组比较,实验组腺实质趋于萎缩。对照组大鼠下颌下腺导管上皮细胞呈瘦素及瘦素受体免疫反应中等阳性,免疫反应物分布于细胞胞质内,细胞核和腺泡细胞呈阴性。4周糖尿病组瘦素及其受体免疫反应呈较强阳性;12周时呈强阳性。结论随糖尿病病程延长,大鼠下颌下腺组织发生退变且瘦素及其受体表达增强,为探讨瘦素变化在糖尿病发病机制中的作用及意义提供了形态学依据。 Objective To study leptin and its receptor expression in submandibular glands of streptozotocin (STZ) induced diabetic rats. Methods 24 male Sprague-Dawley rats were randomly divided into experimental group( 4 and 12 weeks subgroup respectively of 6 rats) and normal control group, per group 12 rats. The diabetic rats model were induced by injection of STZ into peritoneal cavity. After 4 and 12 weeks, rats were killed for assaying blood sugar and weight , and the submandibular glands were observed by HE staining and immunohistochemistry methods. Results Comparing with the control group, the submandibular glandular tissue depauperated in diabetic rats. The leptin and its receptors were expressed in ductal epithelial cells of submandibular glands in three groups. The immunocomplexes distributed in the epithelial cytoplasm. Nuclei and acinus cells were negative. The control group was media and 4 weeks experimental group was higher than that of the control group, while 12 weeks experimental group was higher than that in 4 weeks. Conclusion The submandibular glandular tissue depauperates, expression of leptin and its receptor in submandibular glands gradually increase with diabetes progressing, which provides morphological evidences to further investigate the relationship between leptin and its receptor and the diabetic mechanism.
