摘要 目的研究p27与Mad2蛋白在大肠癌中的表达及其临床意义。方法用原位分子杂交法检测42例大肠癌及正常黏膜中p27mRNA的表达,同时应用免疫组化方法检测相同组织中p27蛋白与Mad2蛋白的表达。结果p27mRNA在正常大肠黏膜及癌组织中的表达阳性率均为100%。p27和Mad2蛋白在大肠癌中的表达阳性率分别为54.76%(23/42)和73.81%(31/42),与正常黏膜中的表达阳性率分别为95.24%(40/42)和50%(21/42)相比,差异均有显著性(P<0.05)。p27蛋白表达与Mad2蛋白表达无相关性(P>0.05),与肿瘤组织分化程度呈负相关(P<0.05),与组织分化、Duke分期等临床病理因素无相关性(P>0.05)。结论p27蛋白与Mad2蛋白表达在大肠癌发生过程中具有重要作用,其检测有助于大肠癌恶性程度评价和预后判断。 Objective To study the expression of p27 and Mad2 protein in colorectal carcinoma and its significances. Methods In situ hybridization for p27 mRNA was performed in 42 colorectal carcinoma and normal mucosa. The expressions of p27 and Mad2 were detected by immunohistochemical staining (S-P method) in the same cases. Results The postive rates of p27 mRNA expression in colorectal carcinoma and normal colorectal mucosa were both 100%. Expressions of p27 and Mad2 protein were 54. 76% and 73.81% respectively in colorectal carcinoma group, and were 95.24% and 50% respectively in normal mucosa. Significant difference of p27 and Mad2 protein expression was found between normal mueosa and carcinoma (P 〈 0. 05 ). No relationship was found between p27 and Mad2 protein expression, p27 protein was inversely correlated with histological grade (P 〈0. 01 ) , but not correlated with other elincopathologieal factors such as histological differetieation and Duke's grade. Conclusion Alterations of p27 and Mad2 protein play an important role in carcinogenesis of eoloreetal carcinoma. Detection of protein expression of p27 and Mad2 assists in evaluating the malignant degree and prognosis of eoloreetal carcinoma.
