苯引起鼠胸腺细胞DNA的损伤及其机制的研究

摘要 目的研究苯对小鼠胸腺细胞DNA的损伤及其机制。方法运用单细胞凝胶电泳技术,将苯分为3个浓度组染毒胸腺细胞,测定胸腺细胞DNA的彗星尾长。结果在有代谢活化系统存在下,苯对小鼠胸腺细胞DNA产生损伤,且呈浓度依赖。但加入抗氧化剂N-乙酰半胱氨酸后,可抑制苯对胸腺细胞DNA的损伤。结论苯能引起胸腺细胞DNA损伤,可能与其代谢物有关。 Objective To detect DNA damage in thymocytes exposed to benzene and explore the mechanism of DNA damage. Methods Alkaline single cell gel electrophoresis (Comet assay) was used to detect the DNA damage. The experiment included three groups according to the concentration of benezene. The grade of tail length of Comet assay was used to assess the DNA damage. Results Benzene could lead thymocytes DNA damage and suggest dose-effect relationships with the existing active metablic system . The DNA damage could be eliminate by added NAC. Conclusion This study indicates that benzene can induce DNA damage of the thymocytes, which might relate to benzene metabolism.
