摘要
建立未衍生化高效液相色谱-蒸发光散射检测器测定茶叶中茶氨酸.采用Polaris-C18(250 mm×4.6 mm)色谱柱,以含0.1%(体积分数)三氟乙酸的水和0.1%(体积分数)三氟乙酸的乙腈作梯度洗脱,流速0.5 ml/min,蒸发光散射检测器为ELSD-2000.茶氨酸浓度在0.062~0.450 mg/ml内,其峰面积的常用对数与进样量的常用对数呈良好的线性关系,平均回收率为97.5%.本方法具有精确、灵敏等特点.
High performance liquid chromatography (HPLC) coupled with evaporative light scattering detector ( ELSD-2000 ) was used for the determination of theanine in tea without derivatization. A Polaris-C18 column (250 mm×4.6 mm) was used with the mobile phase of 0.1%trifluoroacetic acid water solution and 0.1% trifluoroacetic acid acetonitrile solution .The flow rate was at 0.5 ml/min. The linear response ranged from 0.062-0.450 mg/mL for theanine. The recovery was 97.5%. The method was evaluated as accurate and simple.
