摘要
用逆转录-多聚酶链反应(RT-PCR)技术研究了2例正常肾、18例原发性肾癌和1例肾盂癌mdrlmRNA表达状况,结果显示2例正常肾和15例(83.3%)肾癌表达mdrlmRNA基因,另3例肾癌和1例肾盂癌无此基因表达。mdrl基因的表达与肾癌的分级无关(P=0.51)。原发性肾癌mdrl基因的高表达奠定了肾癌内在性耐药的基础。比较不同的基因检测手段显示RT-PCR是简便、快速、灵敏、特异的检测mdrl基因的方法,适于推广。
The expression of mdrl mRNA was studied in 2 normal kidneys,18 renal cell carcinomas(RCC)and 1 transitional cell carcinoma of renal pelvis by reverse transcription-polymerase chain reaction(RT-PCR).The results showed that mdrl gene was expressed in all the 2 normal kidneys and in 83.3%(15/18)of RCC while no mdrl gene was detected in the other 3 RCC and 1 pelvic tumor.No correlation was found statistically between the expression of mdrl gene and the grade of RCCs(P=0.51),The high expression of mdrl gene in primary RCC might be the causative factor of its intrinsic drug resistance.RT-PCR was a sensitive,specific and rapid technique in the detection of mdrl gene for clinical specimens.
