不同细胞因子诱导脐血来源的CIK、NK细胞对K562细胞杀伤活性的研究 被引量:3

摘要 目的:了解不同细胞因子组合扩增的脐血CIK、NK细胞对人K562细胞株的杀伤活性的差异性。方法:通过SCF、FLT3L、IL2、IL7及IL15等细胞因子的不同组合扩增的脐血CIK、NK细胞,分为3组,A组(SCF+IL2+IL7+IL15)、B组(SCF+FLT3L+IL2+IL7+IL15)和C组(IL2+IL7+IL15,对照组);通过MTT法检测各组CIK、NK细胞在不同效靶比下对K562的杀伤率,计算各组的总杀伤单位。结果:经过21天培养A组体系中CIK+NK细胞的比例平均超过60%,B组CIK+NK细胞的比例平均超过70%,而C组约为50%;各组扩增的CBCIKNK细胞同组间在效靶比20∶1时对K562的杀伤率均显著高于10∶1(P<0.01)。在不同效靶比下A组CIKNK细胞对K562的杀伤率显著高于B和C组(P<0.01);C组CIKNK细胞对K562的杀伤率稍高于B组,但两组间对K562的杀伤率无显著性差异。A组总杀伤单位显著高于C组,与B组无显著差异。结论:不同细胞因子组合扩增的脐血CIK、NK细胞对人K562细胞株的杀伤活性有差异,考虑到对效应细胞的扩增效果,B组为具有最佳杀伤活性的脐血CIK、NK细胞培养体系,其中的SCF、FLT3L对CIKNK细胞诱导有协同效果。 Objective: To explore the different cytokine culture system effect on the eytotoxieity of cord blood derived CIK/NK cells against K562 tumor line. Methods: According to the different combination of eytokines, there were three groups, group A( SCF + IL-2 + IL-7 + IL-15) ,group B(SCF+ FLT3L + IL-2 + IL-7 + IL-15) and group C(IL-2 + IL-7 + IL-15,control group). The cytotoxieity of these different expaneled CB-CIK/NK cells against K562 tumor cells were estimated by MTT test and the total lyric unit of each group was calculated. Results: After 21 d in culture, the average percentage of CIK cells plus NK cells in group A exceeded 60%, that of group B exceeded 70%, that of and group C was about 50% .The lytic activity of CB-CIK/NK cells from different group against K562 was higher at 20:1 effeetor-target ratio than that at 10:1 (P 〈 0.01). The eytotoxieity of CB-CAK/NK cells came from group A against K562 line at different effeetor-target ratio signitieantly higher than that of groups B and C( P 〈 0.01 ), the eytotoxieity of CB-CIK/NK cells came from group C against K562 line was sligtly higher than that of groups B, but no significant difference between them. The total Lyric unit(TLu) of group A CB-CIK/NK cells was significantly higher than that of group C , but no significant difference between with group B. Conclusion: The data suggested that there was a difference between the cytotoxicity against K562 of CIK/NK cells generated from different cytokine culture system, concern on the killer proliferation rate, the lytic activity of group B CB-CIK/NK cells is the best.
