男性不育者精液中一氧化氮和顶体酶、透明质酸酶、酸性磷酸酶的变化 被引量:7

摘要 目的探讨男性不育者精液中一氧化氮(NO)和顶体酶(ACE)、透明质酸酶(HYD)、酸性磷酸酶(ACP)的变化。方法参照世界卫生组织(WHO)标准方法进行精液常规分析,按精子密度、活动率不同分组。采用镀铜镉还原荧光法检测NO代谢产物硝酸盐(NO3-);用改良的Kennedy法检测精子ACE活性;用Singer法测定HYD活性;采用磷酸苯二钠法检测ACP。结果72例不育各组NO含量明显高于正常生育组(P<0.01),而ACE、HYD、ACP活性则明显低于正常生育组(P<0.01),随精子密度、活动率减少和畸形率增高而NO含量升高,ACE、HYD、ACP活性降低。结论精液酶类是评价精子成熟、活动率及穿卵受精能力的重要指标,NO对精子酶活性有抑制灭活作用,可影响精子的受精能力,对男性不育的诊断治疗具有重要价值。 Objective To study the change of nitric oxide (NO), acrosome enzyme (ACE), hyaluronidase (HYD) and acid phosphatase (ACP) in human semen. Methods According to the density and activity of the sperm, specimens were divided into 4 groups. The nitrate, a metabolic product of NO was detected by coppered reduced cadmium fluometric assay. The activity of ACE was measured by improved Kennedy assay, HYD activity was measured by Singer assay, and ACP activity was measured by di-sodium phenyl phosphate. Results In 72 infertile men of different groups, the level of NO was significantly higher, and the activity of ACE,HYD and ACP was markedly lower than normal controls. Along with the decrease of density, mobility and the increase of deformity of sperm atozoon, NO became higher and higher,and the activity of ACE,HYD and ACP became lower and lower. Conclusions The determination of ACE, HYD and ACP in human semen is an important index to evaluate the maturity, mobility and fertilization ability of spermatozoon. NO could inhibit the activity of these semen enzymes and may affect the fertilization ability of spermatozoon.
