摘要
目的:采用液相色谱于飞行时间质谱仪联用技术(LC-TOFMS)对中药复方当归补血汤中成分进行分析,并对主要成分进行鉴别。方法:用ZorbaxSBC18色谱柱,以0·1%甲酸水溶液(A)/甲醇(B)梯度洗脱,DAD检测波长280nm,使用ESI离子源,正离子与负离子模式下采集数据。结果:可确定其准确分子量的化合物有27个,能正确鉴定的有15个,另发现7个多肽,给出了其准确分子量。结论:经液相色谱的分离,借助TOFMS测定的准确分子量及其它信息可以鉴别中药复方中的成分,为中药复方中成分的鉴定提供了一种快速准确的方法。
AIM: Apply the techniques of liquid chromatography coupled with time of flight mass spectrometry by using electrspray ionization (LC-ESI/TOFMS) to analyze and identify the compositions of Danggni Buxue (Radix Asteris and Radix Angelicae sinensis) decoction. METHOD: The chromatographic conditions are as follows: Zorbax SB C18(250 mm× 4.6 mm, 5/μm) column; mobile phase: 0.1% formic acid-water (A) and methanol (B), gradient elute; wavelength: 280nm. Use the ESI source and acquire the data in positive and negative mode. RESULT: The accurate molecular weights of twenty-seven compounds were measured in Danggui Buxue decoction. Among them, fifteen compounds were identified. Furthermore, seven polypeptides were detected as well as the accurate molecular weights were measured. CONCLUSION: It is rapid and accurate method that the compositions of compound prescription of Traditional Chinese Medicine can be identified in terms of the separation of liquid chromatography, the accurate molecular weights measured by TOFMS and other information.
