摘要
目的:研究纳米硒多糖的制备及其表征并观察其体外抗CVB3病毒作用.方法:以天然高分子裙带菜多糖为软膜板水相制备纳米硒多糖溶液,并采用瑞利共振散射光谱(RRS)、透射电镜(TEM)、激光散射等手段对产物进行表征.采用结晶紫法考察样品对Hela细胞的毒性作用,及其对CVB3病毒的抑制作用.结果:得到均匀、稳定的球形纳米硒,其平均粒径为 25.3 nm,放置 30 d 后,为 39.1 nm; 由细胞毒性试验得出,各硒糖样品的硒浓度最大不应超过 0.8 μg/ml;硒糖样品的硒浓度在 0.500~0.015 μg/ml 范围内,抗CVB3病毒作用均有一定程度的提高,最大可提高 30% 左右.结论: 将纳米硒多糖开发为一种抗CVB3病毒新药具有广阔的前景.
Objective:To study the synthesis and characterization of selenium nanoparticles with polysaccharides, research its antivirus effects on CVB3 in vitro. Methods:Selenium nanoparticles had been synthesized in an aqueous solution by using polysaccharities from Undaria Pinnatifida( Chary. )Suringer as a soft template. Morphologies of the products were characterized by means of RRS, TEM, and laser scatter. The cytotoxicity of all samples on Hela cells and its antivirus effects on CVB3 in vitro were studied by crystal violet method. Results:Uniform stable selenium nanospheres were obtained, the average particle size of selenium was 25.3 nm, 30 days later, was 39.1 nm, from cytotoxicity test, the max concentration of selenium in samples was 0. 8μg/ml, the antivirus effects on CVB3 in vitro had been improved quite a lot when the concentration of selenium in samples was in the range of 0. 500 - 0. 015μg/ml, the max increase rate of CPE inhibition was about 30%. Conclusion : Nano - selenium polysaccharides has great future to be developed as new medicine against CVB3 virus.
