摘要
抗酒石酸醚性磷酸酶(TRAP)是破骨细胞(OC)的标志酶,为OC所特有。本研究应用改良的VanDeWijngaert和Mostafa方法对培养在骨片上的OC进行TRAP染色,成功地观察了体外培养OC的形态与生存时间。该方法简便、特异性高,能准确计数体外培养的OC数,所用底物为萘酚AS-BI,原料易获得,培养在骨片上的OC生存时间可长达240小时。
Tartrate resistant acid phosphatase(TRAP)is a marker enzyme of osteoclasts(OC)and specific to the OC,With modified Van De Wijngaert and Mostafa's method f the OC cultivated onbone slices was stained with TRAP, Its morphology and survival period was observed successfully.Thernethod was simple,high specific and could be used accurately to number the OC cultivated in vitro,Thesubstrate being used was naphasol AS-BI which could be obtained easily,The OC cultivated on boneslices could survive as long as 240 hours.
