短棒状杆菌有效成分的分离及其作用 被引量:7

摘要 目的减轻短棒状杆菌全菌体制剂接种后的局部疼痛等副作用。方法采用超声破碎、蛋白水解及脱脂的方法提取了细胞壁,通过脾激活试验及抑瘤试验,检测纯化的短棒状杆菌细胞壁活性对家兔股四头肌的局部刺激作用及热原反应。结果纯化的短棒状杆菌细胞壁可使小鼠脾激活指数达到4.8,能抑制艾氏腹水瘤的生长,生存率达到80%;家兔股四头肌肌肉注射后未见局部充血坏死;家兔热原试验未见明显发热。结论纯化的短棒状杆菌细胞壁不仅保留了全菌体制剂所具有的活性,而且明显减轻了局部的刺激作用。 Objective To ease the adverse reaction of whole cell preparation of Corynebacterium parvum. Methods Purify the cell wall of Corynebacterium parvum by ultrasonieafion, hydrolysis of protein and de-fat. Determine the activity of the purified cell wall by splenic lymphocyte activating test and tumor-inhibiting test, study its local stimulating effect on rabbit museulus quadriccps and perform pyrogen test. Results The routine splenic lymphocyte activating index was 4.8. The purified cell wall of Corynebacterium parvum inhibited the growth of Ahrlieh ascites tumor significantly. The survival rate of mice immunized with the purified cell wall was 80%. No local hyperemia or necrosis was observed in the musculus quadriceps of rabbits injected. No significant fever of rabbit was observed in pyrogen test. Conclusion The purified wall of Corynebacterium parvum maintained the activity but eased the adverse reaction of whole cell vaccine of Corynebacterium parvum.
