摘要 目的建立雪莲注射液中绿原酸、芦丁和紫丁香苷的含量测定方法.方法采用Hypersil BDS C-18色谱柱,以乙腈(流动相A)和50 mmol·L-1 KH2PO4缓冲溶液(含体积分数为0.2%的三乙胺,H3PO4调pH=3.6)(流动相B)进行梯度洗脱,流速为1 mL·min-1,检测波长为260 nm.结果样品中绿原酸、紫丁香苷和芦丁的回收率分别为w=99.6%、99.5%和99.0%,RSD分别为0.76%、0.86%和0.96%,绿原酸、紫丁香苷和芦丁的线性分别为0.018~0.360 g·L-1、0.008~0.150 g·L-1和0.007~0.130 g·L-1.结论该方法可用于雪莲及其注射液的质量控制. Objective To develop an RP-HPLC method for the determination of chologenic acid, rutin and syringin in Xuelian(traditional Chinese medicines) injection. Methods A BDS C18 and a gradient elution with acetonitrile-phosphate buffer as the mobile phase were employed. The flow rate was 1 mL· min^- 1 and the detection wavelength was set at 260 nm. Results The linear range of chologenic acid was 0.018 0- 0. 360 g· L^-1 with a regression coefficient of r = 0. 999 9, the average recovery was 99.6 % with RSD of 0.76%;the linear range of syringin was 0.007 5-0.15 g·L^-1 with a regression coefficient of r= 0. 999 9, the average recovery was 99.5 % with RSD of 0.86 % ; the linear range of rutin was 0.006 5- 0. 130 0 g· L^-1 with a regression coefficient of r = 0.999 5, the average recovery was 99.0 % with RSD of 0.96 %. Conclusions This method is proved to be sensitive and accurate, and is suitable for the quality control of Xuelian injection.
