十二元环红霉素衍生物对T淋巴细胞增殖和细胞周期的影响 被引量:2

摘要 目的探讨十二元环红霉素衍生物(LY267108)对T淋巴细胞增殖和细胞周期的影响.方法用MTT法观察LY267108和红霉素对T淋巴细胞增殖的影响;用流式细胞仪经PI染色观察LY267108和红霉素对T淋巴细胞周期的影响.结果LY267108和红霉素抑制T淋巴细胞的增殖IC50值分别为(176.38±10.42)×10-6mol·L-1和(606.28±35.43)×10-6mol·L-1;在3.0~30.0 mg·L-1之间LY267108引起T淋巴细胞G2/M期周期阻滞;红霉素在30.0 mg·L-1和100.0 mg·L-1时出现凋亡峰.结论红霉素衍生物的抗炎活性可能与该类化合物对炎症细胞的影响有关. Objective To study the effects of 12-membered-ring erythromycin derivative LY267108 on the proliferation and cell cycling of T lymphocyte. Method The effects of LY267108 and EM on the proliferation were evaluated by MTT; the effects of LY267108 and EM on celll cycle of T lymphocyte were performed by flow cytometry. Result The proliferation of T lymphocyte were inhibited by LY267108 and erythromycin with IC50(176.38 ± 10.42) × 10^-6mol· L^-1 and (606.28 ± 35.43) × 10^-6mol· L^-1, respective- ly; LY267108 induced a cell cycle arrest in G2/M at 3.0- 30.0 mg· L^- 1, while EM possessed apoptosis-inducing activity at 30.0 mg· L^-1 and 100.0 mg· L^-1. Conclusions The anti-inflammatory activity of erythromycin derivatives may be due partly to their effects on inflammatory cells.
