摘要 为了测量细胞电生理特性和进行药物分析,提出了一种基于光寻址电位传感器(LAPS)的单细胞传感器.用峰峰值为2 mV、频率为1 kHz的正弦信号模拟细胞动作电位,作为传感器的输入信号来观测系统的灵敏度;采用涂敷法和改变器件表面粗糙度的方法,将细胞固定在器件表面以改善传感器灵敏度;选择1μm/mL、10μg/mL两种质量浓度乙酰胆咸(Ach)作为大脑皮层神经细胞的刺激药物,检测在不同浓度药物作用下神经细胞的动作电位.实验结果表明,涂敷法固定细胞提高了传感器的灵敏度,该细胞传感器能够检测出模拟动作电位,并能够检测出不同浓度的乙酰胆碱. To measure neural electrophysiological characteristics and to evaluate drugs, a novel cell-based biosensor based on light-addressable potentiometric sensor (LAPS) was proposed. The sensor sensitivity was detected by adding a sine wave signal with amplitude of 2 mV and frequency of 1 kHz as simulated neural action potential (AP). Neurons were immobilized on the sensor surface by depositing bio-layersand changing surface roughness to improve the sensitivity. Acetylcholine (Ach) with concentration of 1 mg/L, 10 mg/L was chosen as stimulating drug of cortical neuron, and the AP of the neuron was detected under different drug concentrations. The results show that surface modification can improve the sensor sensitivity, and the proposed sensor can successfully detect the simulated AP signal. The single-cell sensor is able to distinguish the response of neurons under different drug concentrations.
