src基因产物在贲门癌中表达的免疫组织化学观察

An Immunohistochemical Observation of src Gene Product in Cardiac Carcinoma

用ｃ－ｓｒｃ基因产物ＰＰ６０ｃ－ｓｒｃ的特异性单克隆抗体Ｍａｂ３２７对５６例贲门癌、３６例癌旁组织及２０例淋巴结转移癌进行免疫组织化学研究。结果，癌旁正常上皮，增生上皮、腺癌及淋巴结转移癌ＰＰ６０ｃ－ｓｒｃ阳性率分别为２９．４％（１０／３４）、９４．４％（３４／３６）、７１．４％（４０／５６）、６０．０％（１２／２０），几者间有显著性差异（Ｐ＜０．００５）。腺癌及癌旁增生上皮ＰＰ６０ｃ－ｓｒｃ表达量显著高于癌旁正常上皮（Ｐ＜０．００５）。乳头状、管状、低分化及粘液腺癌ＰＰ６０ｃ－ｓｒｃ阳性率分别为７５．０％（６／８）、８１．８％（１８／２２）、５０．０％（１０／２０）、１００．０％（６／６），几者间有显著性差异（Ｐ＜０．０５），其中粘液、管状腺癌的表达量高于乳头状和低分化腺癌（Ｐ＜０．００５）。结果提示，ｃ－ｓｒｃ基因激活表达与贲门癌发生发展有关，在其癌变过程中ＰＰ６０ｃ－ｓｒｃ表达量升高，并与贲门癌的组织学类型、分化和转移有关。

In this experiment, 56 cases of cardiac carcinomas, 36 cases of tissues adjacent to carcinoma and 20 cases of metastatic lymph nodes were studied immunohistochemically by using specific monoclonal antibody of c-src gene product (pp60c-src), namely Mab 327. The positive rates of pp60c-src in the normal epithelia, proliferative epithelia adjacent to carcinoma, cardiac adenocarcinoma and metastatic lymph nodes were 29.4%(10/34), 94.4%(36/34), 71.4%(40/56), and 60.0%(12/20), respectively. There were statistically significant difference in them (P<0. 005). The amount of pp60c-src in the adenocarcinomas and proliferative epithelia adjacent to carcinoma were significantly higher than those in the normal epithelia adjacent to carcinoma (P<0.005).The positive rates of pp60c-src of cardiac adenocarcinomas with different histological types were as follows: 75.0%(6/8) for the papillary type, 81. 8%(18/22) for the tubal type, 50. 0%(10/20) for the low differentiated type, and 100. 0% (6/6) for the mucous type, the difference of the positive rates were significant statistically (P<0.05).And the quantity of the gene expression in the mucous and tubal adenocarcinomas were significantly higher than those in the papillary and low differentiated types(P<0. 005). It is suggested that the activity and express of c-src gene was closely associated with the initiation and development of the cardiac adenocarcinoma.
