摘要
应用聚合酶链反应(PCR)技术和寡聚核甘酸探针杂交法检测膀胱癌中Ha-ras癌基因第12位密码子G→T突变,同时应用免疫组织化学方法检测了rasPZI在膀胱癌中的表达。在29例膀胱癌中有10例存在Ha-ras癌基因第12位密码子点突变,突变率为34.5%,有18例rasP21表达为阳性,阳性率为62.1%。结果提示Ha-ras癌基因第12位密码子点突变和rasP21的表达与膀胱癌病理分级、临床分期和膀胱癌的复发密切相关。Ha-ras癌基因第12位密码子点突变与rasP21的表达是一致的。
G→T mutation on codon 12 of Ha-ras oncogene was detected in bladder cancer by means of polymerase chain reaction (PCR) technics and blothy bridization assays with specific oligonucleotide probe. The expression of ras P21 was investigated by immunohistochemical techniec.In 10 of 29 bladder cancer specimens point mutation in codon 12 of Ha-ras oncogene was found,with a mutation rate of 34.5%.The P21 was positive in 18,with a positive rate of 62. 1%.The results showed that there was relationship between the point mutation of Ha-ras oncogene and ras P21 expression and the pathological grade,clinical stages and recurrence of tumor.The expression of ras P21 was consistent with point mutation in codon 12 of Ha-ras oncogene.
