中缝大核对急性心肌梗塞大鼠交感心血管反应的影响

摘要 目的:观察中缝大核对急性心肌梗塞大鼠交感心血管反应的影响。方法:SD雄性大鼠60只,随机分为6组:正常对照组,急性心梗对照组,生理盐水注射组,6-OHDA注射组,小剂量谷氨酸注射组,大剂量谷氨酸注射组。各组均取第3胸髓节段和左心室置于4%多聚甲醛固定24h,切片,片厚5μm。1套行免疫组织化学染色;1套行尼氏染色。左心室切片行HE染色。结果:6-OHDA注射组和生理盐水注射组阳性反应神经元数量与急性心梗对照组比较无显著差异;正常对照组几乎未见阳性反应神经元;小剂量谷氨酸注射组阳性反应神经元数量增多,与急性心梗对照组比较差异显著;大剂量注射组阳性表达减少,与急性心梗对照组比较差异显著。结论:中缝大核对急性心肌梗塞大鼠交感心血管反应有影响。 Objective: To observe the effect of nucleus raphe magnus on sympathetic cardiovascular response of rat following acute myocardial infarction(AMI). Methods: 60 adult male Sprague-Dawley rats were divided randomly into six groups, normal control group, acute myocardial infarction control group, physiological saline injection group, 6-OHDA injection group, lesser doses of glutamate injection group and greater doses of glutamate injection group. T3 spinal cords and left ventricles of all rats were removed and placed in 4% paraformaldehyde solution for 24h,then the specimens were cut for serial 5μm transverse sections. The slices were dyed with immunohistochemistry staining and Nissl's staining respectively. The sections of left ventricles were stained with H.E method. Results: The number of positive neurons had no significant difference between 6-OHDA injection group,physiological saline injection group and acute myocardial infarction control group. There was almost no positive neuron in normal control group. Compared with the acute myocardial infarction control group, injection of lesser doses of glutamate significantly increased the number of positive neurons,while greater doses of glutamate significantly reduced it. Conclusion: Nucleus raphe magnus had a influence on sympathetic cardiovascular response of rat following acute myocardial infarction.
