摘要
目的:从牛肌腱中提取I型胶原,制作适宜于人工骨材料的基质支架。方法:酸溶法提取牛肌腱中的胶原蛋白,并经盐析、透析提纯,用紫外分光光度法、氨基酸分析、常规组织切片、扫描电镜对提纯的胶原进行检测。冷冻干燥法获得多孔胶原基质支架。结果:提纯精制的胶原蛋白符合I型胶原蛋白的特征。胶原支架块具有疏松多孔的三维结构,孔隙率90.0%以上。结论:提取的胶原符合I型胶原的特点,适宜于制作成为人工骨生物材料的基质支架。
Type Ⅰ collagen was extracted and purified from bovine tendon, then made to base matrix scaffold of artificail bone. Methods: Raw collagen was extracted from bovine tendon in acetic acid, then by salting out, dialysis and lyophilization , pure collagen scaffold was acquired. Ultraviolet spectrophotography , amino acid analysis ,histology observtion , scanning electron microscope were used to evaluate the pure collagen. Results: The purified collagen was coincidence with type Ⅰ collagen. Collagen matrix scaffold was similar to sponge , the ratio of porosity was above 90%. Conclusion:Collagen matrix scaffold had suitable strcture to form base scaf- fold of artificial bone.
