肿瘤全溶物冲击的DC联合超抗原SEB对效应细胞免疫活性的影响

摘要 目的:探讨肿瘤全溶物冲击致敏的树突状细胞(DC)联合金黄色葡萄球菌肠毒素B(SEB)对效应淋巴细胞免疫活性的影响。方法:以DC或联合SEB刺激效应细胞,流式细胞术(FCM)检测产生TNF-α的效应细胞阳性率,MTT法检测效应细胞的增殖效应,以及HcaF肝癌细胞全溶物冲击后的DC联合SEB刺激T细胞时对HcaF细胞的杀伤效果。结果:100ng/mL SEB刺激效应细胞5 d时产生TNF-α分泌高峰,阳性细胞率为(50.17±2.23)%,而联合DC后(DC∶T=1∶30),5 d时TNF-α分泌峰值更高,阳性细胞率为(60.74±2.76)%,两组比较差异有统计学意义,P=0.002;效应细胞增值指数(SI)在联合两者时为2.67±0.47,高于单独SEB处理的2.58±0.38;肿瘤全溶物单纯冲击效应细胞前后对HcaF的杀伤率差异无统计学意义,联合SEB后,杀伤率由无SEB时的(42.92±7.84)%提高到(59.94±3.83)%,而进一步在DC辅助冲击后的杀伤率(62.44±6.62)%更高于冲击前的(52.33±7.02)%,两组比较差异有统计学意义,P=0.001。结论:肿瘤全溶物冲击的DC联合SEB能诱导效应细胞明显增值、活化,并产生高效的抗肿瘤效果。 OBJECTIVE:To investigate the influence of den dritic cells (DC) pulsed with whole tumor lysates (WTL) combined with staphylococcal enterotoxin B (SEB) eliciting the effector ceils immune activity. METHODS: Stimulating the effector ceils only by DC or combined with SEB,the expression rate of effector ceils excreting tumor necrosis factor α (TNFα) was detected by FCM, the effector ceils proliferation and antitumor effect stimulated only by dendritic ceils pulsed with WTL from mouse hepatocarcinoma HcaF cells or combined with SEB were detected by MTT assay. RESULTS:There was a peak of TNFα stimulated by 100 ng/mL SEB, and the TNFα excretion rate of effector cell was (50. 17 ± 2.23) %, lower than the rate (60. 74± 2.76)% produced by SEB combined with DC (DC : T= 1 : 30) at the same time, P= 0. 002; the stimulating index (SI) of effector cells treated by SEB with DC was 2.67±0.47, higher than that only treated by SEB, 2.58±0.38;the antiHcaF effects produced by effector cell treated only with or without WTL was of no signification; combined with SEB, the antiHcaF rate was (59.94 ± 3.83 ) %, higher than the rate without SEB, (42.92±7.84) % ; further plused with DC, the anti HcaF rate was (62.44 ± 6.62)%, higher than the rate without DC, (52.33± 7.02)%, P: 0.001. CONCLUSION:DC pulsed with WTL combined with SEB induces effector cell effective proliferation, activation and high antitumor efficiency, and shows a potential kind of operative antitumor immunotherapy.
