摘要
目的:探讨肺腺癌组织中血管内皮生长因子(vascular endothelial growth fac-tor,VEGF)mRNA的表达和外周血微转移的关系。方法:应用逆转录-聚合酶链式反应(RT-PCR)方法,检测33例肺腺癌组织中VEGF mRNA的表达。以CK19为标志物,以RT-PCR检测33例肺腺癌患者和10位正常人外周血的微转移。结果:33例肺腺癌患者中,VEGF mRNA的阳性率为78·79%(26/33),外周血CK19的阳性率为57·58%(19/33),10位正常人外周血的CK19的阳性率为0。肺癌组织中,VEGF mRNA的表达和腺癌的淋巴结转移及病理分期密切相关,P值分别为0·002和0·008;VEGF mR-NA阳性表达组和VEGF mRNA阴性表达组的CK19的阳性率差异有统计学意义,P=0·009。结论:肺腺癌患者组织中的VEGF mRNA表达和淋巴结及血行转移关系密切,联合检测肺腺癌组织中的VEGFmRNA的表达和外周血微转移不仅能够评估肺癌侵袭转移的程度,而且对正确评价肺癌的病理分期、预后及制定合理的治疗方案有重要意义。肿瘤防治杂志,2005,12(19)
OBJECTIVE: To evaluate the relationship between micrometastasis and vascular endothelial growth factor (VEGF) mRNA in lung adenocarcinoma. METHODS: VEGF mRNA and CK19 as the markers of disseminated tumor cells were detected by the method of reverse transcripase polymerase chain reaction in 33 cases of lung adenocarcinoma and 10 of normal control subjects. RESULTS: The positive rate of VEGF mRNA in 33 cases of lung adenocarcinoma was 78. 79% (26/33); the positive rate of CK19 mRNA in peripheral blood of 33 cases of lung adenocarcinoma was 57.8%(19/33) ; the positive rate of normal control subjects was 0. The expression of VEGF mRNA was significantly correlated with lung adenocarcinoma lymph node metastasis and TNM stage, P = 0. 002, P = 0. 008. The relationship between VEGF mRNA expression and the micrometastasis in peripheral blood was significant. The preoperation expression of CK19 mRNA in peripheral blood with VEGF mRNA positive group was significantly higher than that of VEGF mRNA negative group, P= 0. 009. CONCLUSIONS: The expression of VEGF mRNA in lung adenocarcinoma is significantly correlated with lymph node metastasis and hematogenous tumor metastasis. Examination of VEGF mRNA and the circular micrometastasis can evaluate the intensity of invasion and metastasis and can chose the reasonable treatment.
