Study Advance in Rhododendron Tissue Culture

杜鹃组织培养以茎尖、带腋芽的幼茎、幼叶、花雷、种子等作外植体;以浓度0.1%HgCl2消毒5～8min,或浓度0.2%HgCl2消毒2～3min;以Read、Mccown、Anderson、1/4 MS、1/10MS、MS、改良MS、ER等为基本培养基;以ZT2.0～5.0 mg/L或2ip10～15 mg/L或KT2.0～5.0mg/L,配合NAA或IAA 0.05～0.5 mg/L诱导不定芽及愈伤组织;以IBA或NAA为0.5～1.0 mg/L+活性炭0～0.3%诱导根形成;以浓度3%的蔗糖作碳源;以AC(活性炭)0.1%、PVP(聚乙烯吡咯烷酮)100 mg/L控制褐化;pH值为5.0～5.4;光照强度为1 500～2 500 lx,光照时间为16h/d,黑暗8h/d,培养温度(25+2)℃.

The Rhododendron shoot tip, cladium, delicate leaf, alabastrum and seed are usually used as explants in tissue culture. 0.1% HgCl2 or 0.2% HgCl2 was the best disinfecting liquid. The best medium was Read, Mccown, Anderson, 1/4 MS, 1/10MS, MS and ER, and the best optimal hormone cornponent and concentration were 2.0 - 5.0 mg/L ZT, 10 - 15 mg/L 2ip or 2.0 - 5.0 mg/L KT and 0.05 - 0.5 mg/L NAA or IAA in induction culture for callus and adventitious bud. 0.5 - 1.0 mg/L IBA or NAA and 0 - 0.3% AC was the best in root formation.3% sugar was the best carbon origin .0.1%AC and 100 mg/L PVP,pH value = 5.0 - 5.4 1 500 - 2 500 lx for 16 h/d and （25 ＋ 2）℃ was beneficial to healthy development.
