摘要
柔红霉素(Daunomycin,DNR)是一种广谱抗癌药物。由于它对心肌、骨髓等正常组织有毒性作用而常常发生严重的毒副反应,使临床应用受到很大限制。应用抗肿瘤单克隆抗体与抗癌药交联制备免疫抗癌药,不仅可提高肿瘤组织中的药物浓度,且可减少毒副反应。
Daunomycin (DNR), an anthracycline anticancer. drug, was covalently attachedvia a dextran brige to the anti-human gastric cancer monoclonal antibody (MG^7).The molar ratio of MG^7: Dextram: DNR was 1: 2.5: 60 in the conjugate. Immunofluo-rescent assay revealed that the immunological activity of the antibody in the DNR-Dextran-MG^7 conjugate was retained substantially. In vitro, antitumor activity ofDNR-Dextran-MG^7 conjugate against gastric carcinoma cell line KATO Ⅲ was foundto be similar to that of free DNR at low concentration but stronger at higher cencen-tration and exert lower cytotoxic effect on unrelated cell Hep-2 (human laryngo-carciroma cell line).
