板桥党参多糖的分离纯化及组成研究 被引量:20

摘要 目的从板桥党参中分离纯化板桥党参多糖(COP),并研究其组成。方法采用水提醇沉的方法从板桥党参中提出粗COP,并将其脱蛋白质后,用多种有机溶刑洗涤,得精制COP。然后采用DEAE-纤维素柱分离纯化精制COP,采用体积排阻色谱-多角度激光光散射法(SEC-LLS)对其相对分子质量进行测定,并用气相色谱法对其组成进行研究。结果分离得到COP-Ⅰ和COP-Ⅱ两种水溶性多糖,其重均相对分子质量分别是6 308和7 086。其中COP-Ⅰ由甘露醇、果糖、葡萄糖组成,比例为1: 0.05:1.56,咔唑反应呈阴性;COP-Ⅱ由甘露醇、果糖、葡萄糖、半乳糖组成,比例为1:0.11:1.07:0.37,咔唑反应呈阳性。结论COP-Ⅰ是一种中性杂多糖,COP-Ⅱ是一种酸性杂多糖。 OBJECTIVE To extract and purify Codonopsis tangshen Oijv. polysaccharides(COP) from Codonopsis tangshen Oijv., to analyze their composition. METHOIS After extracted with hot water and precipitated with alcohol, the crude COP was separated from Codonopsis tangshen Oijv.. After being removed protein and washed by organic solvents, the refined COP was obtained. Refined COP was iso- lated and purified by DEAE-cellulose column. And their molecular weights and chemical components were measured by SEL-LLS and gas chromatography, respectively.RESULTS The molecular weights of COP-Ⅰand COP-Ⅱ were 6 308 and 7 086, respectively. COP-Ⅰ contained mannose, fructose and glucose in the mole ratio of 1:0.05: 1.56, the carbazole reaction was negative. COP-Ⅱ contained mannose, fructose, glucose and galactose in the mole ratio of 1 : 0.11 : 1.07 : 0.37, the carbazole reaction was positive. CONCLUSION COP-Ⅰ is a kind of neutral heteropolysaccharide, and COP- Ⅱ is a kind of acidic heteropolysaccharide.
