黏附分子T-cadherin表达对C6细胞恶性特性的影响 被引量:6

Effect of T-cadherin gene expression on malignant biological characteristics in C6 glioma cells

摘要 目的探讨Tcadherin分子表达对胶质母细胞瘤C6细胞的恶性生物学特征的影响。方法C6细胞分别转染pcDNA3.Tcadherin和pcDNA3表达质粒后,筛选3个表达不同水平Tcadherin的C6克隆和2个不表达Tcadherin的C6克隆,研究Tcadherin分子表达水平对C6细胞的细胞黏附、迁移及聚集能力的影响。结果与不表达Tcadherin的C6细胞相比,表达Tcadherin的C6细胞在附着后1h和4h时的细胞黏附能力均显著增强,分别增强61.2%和25.1%(P<0.01),而其细胞迁移能力平均下降72.6%(P<0.010)。同时,Tcadherin表达诱导显著的同源细胞聚集,聚集指数平均下降52.4%(P<0.01)。结论Tcadherin分子表达显著抑制胶质母细胞瘤C6细胞的恶性生物学特性。 Objective To study “he effect of T-cadherin gene expression on malignant biological characteristics in C6 glioma cells. Methods Three C6 clones with different expression levels of T-cadherin and two C6 clones without T-cadherin expression after transfecting C6 cells with pcDNA3. T-cadherin and pcDNA3 using lipofectin were chosen to study the effects of T-cadherin gene expression on cell adhesion, motility and aggregation. Results Compared to the non-T-cadherln-expressing C6 cells, T-caderin-expressing C6 cells indicated enhanced ceil adhesion, and average 61.2% and 25.1% increases in cell adhesion at 1 and 4 h culture were observed (P〈0.01), but their motility was decreased by 72.6% ( P 〈 0.01 ). At the same time, T-cadherin-expressing C6 cells demonstrated significantly homophilic aggregation, and aggregation index was decreased by 52.4 % compared to the non-T-cadherin-expressing C6 cells ( P 〈 0.01 ). Conclusion T-cadherin gene expression significantly suppresses the malignant biological characteristics of C6 glioma cells.
